- Hoshikawa Bianca (星川 ビアンカ Hoshikawa bianka?) / Cure Lila (キュア リラ Kyuarira?)
The lead cure of the series. Bianca is a passionate and energetic young 13-year old with dreams of becoming a famous singer. Her Pretty Cure alter ego is Cure Lila, the Legendary Floret of purity whose theme colour is white. - Kashikoi Akari (賢い 明り Kashikoi akari?) / Cure Iris (キュア アイリス Kyuaairisu?)
Shy but intelligent, Akari is the student librarian of Hikariyama Academy's library and, as well as that, she can get startled easily. Her Pretty Cure alter ego is Cure Iris, the Legendary Floret of Wisdom whose theme colour is blue. - Kiiro Sayaka (黄色さやか Kiiro sayaka?) / Cure Plumeria (キュアプルメリア Curepurumeria?)
A transfer student from Kyoto, Sayaka is very fashionable and optimistic, almost getting excited about everything. Her Pretty Cure alter ego is Cure Plumeria, the Legendary Floret of energy whose theme colour is yellow. - Shigaisen Viola (紫外線ビオラ Shigaisen biora?) / Cure Nepeta (キュアネペタ Kyuanepeta?)
A girl from the Fiore Kingdom, Viola is an intelligent and kind girl. Despite this, she can get very strict if someone is very irresponsible. Her Pretty Cure alter ego is Cure Nepeta, the Legendary Floret of kindness whose theme colour is purple. - Awabeni Rose (淡紅ローズ Awabeni rōzu?) / Cure Hibiscus (キュアハイビスカス Kyuahaibisukasu?)
A young girl with a passion for drawing, Rose can draw anything. Her Pretty Cure alter ego is Cure Hibiscus, the Legendary Floret of Creativity whose theme colour is pink.

- Daylily (デイリーリリー Deirīrirī?)
The main mascot of the series. Daylily is sweet and kind but can sometimes be quite stubborn.
- Senbonzakura (千本桜 Senbonzakura?)
The queen of the Fiore Kingdom. Senbonzakura is very protective of her kingdom and family even protecting her new-born daughter 13-years ago.
